Increased resistance to rice stem borers in Indonesian rice cultivars (Rojolele cv) containing cry IAb gene

Rice stem borers is one of the main pests in rice plantations in Indonesia. The absence of a source of resistance genes against rice stem borer from rice plants, made it necessary to assemble rice (Oryza sativa) cv Rojolele plants resistant to the rice yellow stem borer (Scirpophaga incertulas). The aim of this research was to find out the effect of cryIAb insertion on Indonesian rice cultivar resistance to yellow stem borer. The approach was by constructing a binary vectors containing two separate T-DNA’s, one containing a drug resistance selection marker gene (hpt) and the other one containing cryIAb. Transformation with pARBIO resulted in 21 transgenic plants having the cry1Ab. Analyses of the integration patterns of the transgenes using cryIAb probe in transgenic plant showed single copy insertions. Bioassay of fourth generation (T3) free selectable marker transgenic plants showed resistant improvement against yellow stem borer than non-transgenic control plants.
